High Levels Of A Protein Linked To Increased Stroke Risk
NEW ORLEANS (Dow Jones)—A relatively unknown protein has been shown to sharply elevate the risk of strokes even among people who have healthy blood cholesterol levels.
A new analysis of data from an ongoing National of Institutes of Health clinical trial of 15,000 people that is studying various cardiovascular risks was presented Tuesday at the American Heart Association’s annual meeting here.
The study’s lead investigator, Dr. Christie M. Ballantyne, the director of cardiovascular disease prevention at Baylor University, focused on a protein called Lp-PLA2. That protein appears to be an independent risk factor for developing cardiovascular disease outside of other well-known factors such as having high levels of “bad,” or LDL, cholesterol and low levels of HDL, or “good,” cholesterol.
He said that people who have high levels of Lp-PLA2 have an 11-fold higher risk of having a stroke as those with a low level of the protein. Recently, another study that followed European men over a 14-year period found that those with elevated levels of Lp-PLA2 have a 37% greater risk of having a coronary event such as a heart attack.
GlaxoSmithKline PLC (GSK) is developing a drug aimed at reducing Lp-PLA2 levels. The firm is studying various compounds.
Preliminary data from one such compound - 480848 - also were released at the AHA meeting Tuesday. The compound was studied in 103 patients, all of whom had significant plaque buildup in their carotid artery, the main artery in the neck. When the plaque breaks off it can clog a blood vessel and cause a stroke.
The patients were divided into three groups. One group received 40 milligrams of the compound, while others received 80 milligrams daily for an average of 14 days. A third group received a placebo.
The results showed that Lp-PLA2 activity in the plaque was reduced by 80% in the people taking the higher dose and by 52% in those on the lower dose. Lp-PLA2 is believed to have a role in the formation of plaque in the arteries.
Dr. Lawson Macartney, GlaxoSmithKline’s senior vice president for cardiovascular drug development, said in an interview that the compound is being tested in larger groups of patients. He said it will be at least five years before the firm has enough data to approach the Food and Drug Administration for a drug approval.
He said he believes that Lp-PLA2 is like cholesterol was in the 1980s, when its links to cardiovascular disease were not well-understood.
